Tujuan penelitian ini adalah untuk mengetahui: (1) Proses pelaksanaan pembelajaran Lukis Totebag di kelas X MIA 3 SMA Negeri 3 Boyolali tahun ajaran 2017/2018. Dan (2) Bagaimana bentuk hasil karya Lukis Totebag yang dihasilkan siswa di kelas X MIA 3 SMA Negeri 3 Boyolali tahun ajaran 2017/2018. Penelitian ini menggunakan pendekatan kualitatif. Sumber data yang digunakan adalah informan yang dipilih yaitu Bapak Subandiyo S.Pd selaku guru mata pelajaran seni budaya di kelas X MIA 3 SMA Negeri 3 Boyolali, serta foto proses pembelajaran, hasil karya siswa dan dokumen arsip. Teknik yang digunakan dalam pengumpulan data adalah observasi langsung, wawancara terstruktur dan mendalam, serta dokumentasi. Uji validitas data dilakukan dengan membandingkan sumber data yang di peroleh berupa daftar hasil wawancara dengan Bapak Subandiyo S.Pd selaku guru mata pelajaran Seni Budaya dengan siswa di kelas X MIA 3 SMA Negeri 3 Boyolali, serta review informant. Analisis data yang digunakan adalah model analisis mengalir, yaitu: reduksi data, sajian data, dan penarikan kesimpulan. Hasil penelitian ini menunjukkan bahwa: (1) Pembelajaran Lukis Totebag diawali dengan pembuatan RPP, selanjutnya pembelajaran dilaksanakan selama tiga kali pertemuan. Strategi yang digunakan guru dalam pembelajaran ini adalah pendekatan scientific. Metode pembelajaran yang digunakan meliputi metode ceramah, tanya jawab, diskusi, dan pemberian tugas. Media pembelajaran yang digunakan berupa slide power point dan media visual berupa sampel karya dari guru. Evaluasi pembelajaran dilakukan dengan menilai aspek kognitif, afektif, dan psikomotorik. Proses pembuatan karya dilakukan dengan beberapa langkah, yaitu membuat sketsa, proses pewarnaan, dan finishing. (2) Secara umum pembuatan karya lukis totebag siswa sudah baik, teknik lukis pada pewarnaan dan finishing dalam membuat karya lukis totebag sudah baik. Karya lukis totebag yang dihasilkan oleh siswa sudah mengandung unsur-unsur seni rupa, yaitu: garis, bentuk, bidang, gelap terang, dan warna. Selain itu, karya lukis totebag yang dihasilkan oleh siswa juga sudah mengandung prinsip-prinsip seni rupa, yaitu: irama (rhytm), dominasi (dominance), keseimbangan (balance), kesatuan (unity), keserasian (harmony), dan kesebandingan (proportion).     Chemical Characterization and Hypoglycaemic Activities In Vitro of Two Polysaccharides from <i>Inonotus obliquus</i> by Submerged Culture

    Polysaccharides from the fungus Inonotus obliquus have been found to be biologically active. In this study, we carried out a preliminary characterisation and assessment of the hypoglycaemic activities of the polysaccharides (IOEP) from Inonotus obliquus obtained by liquid fermentation. Two polysaccharides, IOEP1 and IOEP2, were isolated from IOEP. IOEP1, with a molecular weight of 20 KDa, was mainly composed of galatose and mannose, while IOEP2, with a molecular weight of 200 KDa, was mainly composed of arabinose. Fourier-transform infrared analysis showed that both IOEP1 and IOEP2 were pyran-type polysaccharides. 1H-NMR spectra showed that the glycosidic bonds of IOEP1 and IOEP2 were both &#945;-type and &#946;-type. In addition, IOEP1 and IOEP2 strongly increased the glucose consumption of HepG2 cells and insulin-resistant HepG2 cells in vitro. These findings provide a theoretical basis that IOEP1 and IOEP2 might be suitable as anti-diabetes agents in functional foods and natural drugs

    Enhanced Enzymatic Hydrolysis and Structural Features of Corn Stover by NaOH and Ozone Combined Pretreatment

    A two-step pretreatment using NaOH and ozone was performed to improve the enzymatic hydrolysis, compositions and structural characteristics of corn stover. Comparison between the unpretreated and pretreated corn stover was also made to illustrate the mechanism of the combined pretreatment. A pretreatment with 2% (w/w) NaOH at 80 &deg;C for 2 h followed by ozone treatment for 25 min with an initial pH 9 was found to be the optimal procedure and the maximum efficiency (91.73%) of cellulose enzymatic hydrolysis was achieved. Furthermore, microscopic observation of changes in the surface structure of the samples showed that holes were formed and lignin and hemicellulose were partially dissolved and removed. X-ray Diffraction (XRD), Fourier Transform Infrared Spectroscopy (FTIR) and Cross-Polarization Magic Angle Spinning Carbon-13 Nuclear Magnetic Resonance (CP/MAS 13C-NMR) were also used to characterize the chemical structural changes after the combined pretreatment. The results were as follows: part of the cellulose I structure was destroyed and then reformed into cellulose III, the cellulose crystal indices were also changed; a wider space between the crystal layer was observed; disruption of hydrogen bonds in cellulose and disruption of ester bonds in hemicellulose; cleavage of bonds linkage in lignin-carbohydrate complexes; removal of methoxy in lignin and hemicellulose. As a result, all these changes effectively reduced recalcitrance of corn stover and promoted subsequent enzymatic hydrolysis of cellulose

    Structure Characterization and Hypoglycaemic Activities of Two Polysaccharides from Inonotus obliquus

    In the present study, two polysaccharides (HIOP1-S and HIOP2-S) were isolated and purified from Inonotus obliquus using DEAE-52 cellulose and Sephadex G-100 column chromatography. The structural characterization and in vitro and in vivo hypoglycaemic activities of these molecules were investigated. HPLC analysis HIOP1-S was a heterpolysaccharide with glucose and galactose as the main compontent monosaccharides (50.247%, molar percentages). However, HIOP2-S was a heterpolysaccharide with glucose as the main monosaccharide (49.881%, molar percentages). The average molecular weights of HIOP1-S and HIOP2-S were 13.6 KDa and 15.2 KDa, respectively. The &beta;-type glycosidic bond in HIOP1-S and HIOP2-S was determined using infrared analysis. 1H-NMR spectra indicated that HIOP2-S contains the &beta;-configuration glycosidic bond, and the glycoside bonds of HIOP1-S are both &alpha;-type and &beta;-type. The ultraviolet scanning showed that both HIOP1-S and HIOP2-S contained a certain amount of binding protein. Congo red test showed that HIOP1-S and HIOP2-S could form a regular ordered triple helix structure in the neutral and weakly alkaline range. HIOP1-S and HIOP2-S showed strong &alpha;-glucosidase inhibitory activities and increased the glucose consumption of HepG2 cells. In addition, Streptozotocin (STZ)-induced hyperglycaemic mice were used to evaluate the antihyperglycaemic effects of HIOP1-S and HIOP2-S in vivo. The results showed that HIOP2-S had antihyperglycaemic effects. Taken together, these results suggest that HIOP1-S and HIOP2-S have potential anti-diabetic effects

    Simultaneous Use of Stimulatory Agents to Enhance the Production and Hypoglycaemic Activity of Polysaccharides from Inonotus obliquus by Submerged Fermentation

    This study aimed to determine the effect of applying stimulatory agents to liquid cultured Inonotus obliquus on the simultaneous accumulation of exo-polysaccharides (EPS) and their monosaccharide composition. Different stimulatory agents (VB6, VB1, betulin and birch extract) were investigated for their effects on active exo-polysaccharides by submerged fermentation of I. obliquus. The mycelial biomass, reducing sugar content, EPS yield and &alpha;-glucosidase inhibition rate were determined, and the EPS obtained was analyzed for monosaccharide composition. The results showed that the addition of all the four stimulatory agents could significantly increase the inhibitory activity against &alpha;-glucosidase of EPS than the control, whereas EPS from 4 &mu;g/mL VB1-containing medium had the best effect with an estimated IC50 value 24.34 &mu;g/mL. Among the four stimulatory agents, VB6 gave maximum production of mycelial biomass and EPS at the concentration of 4 &mu;g/mL with a increase of 50.79% and 114.46%, respectively. In addition, betulin had a significant effect on increasing the EPS yield and activity, and birch extract had a significantly stimulatory effect on the mycelial growth and the polysaccharides activity, only slightly worse than VB6 and VB1. Moreover, the addition of different stimulatory agents changed the monosaccharide composition of polysaccharides, which had a correlation with polysaccharide activity

    Emerging biomaterials for tumor immunotherapy

    Abstract Background The immune system interacts with cancer cells in various intricate ways that can protect the individual from overproliferation of cancer cells; however, these interactions can also lead to malignancy. There has been a dramatic increase in the application of cancer immunotherapy in the last decade. However, low immunogenicity, poor specificity, weak presentation efficiency, and off-target side effects still limit its widespread application. Fortunately, advanced biomaterials effectively contribute immunotherapy and play an important role in cancer treatment, making it a research hotspot in the biomedical field. Main body This review discusses immunotherapies and the development of related biomaterials for application in the field. The review first summarizes the various types of tumor immunotherapy applicable in clinical practice as well as their underlying mechanisms. Further, it focuses on the types of biomaterials applied in immunotherapy and related research on metal nanomaterials, silicon nanoparticles, carbon nanotubes, polymer nanoparticles, and cell membrane nanocarriers. Moreover, we introduce the preparation and processing technologies of these biomaterials (liposomes, microspheres, microneedles, and hydrogels) and summarize their mechanisms when applied to tumor immunotherapy. Finally, we discuss future advancements and shortcomings related to the application of biomaterials in tumor immunotherapy. Conclusion Research on biomaterial-based tumor immunotherapy is booming; however, several challenges remain to be overcome to transition from experimental research to clinical application. Biomaterials have been optimized continuously and nanotechnology has achieved continuous progression, ensuring the development of more efficient biomaterials, thereby providing a platform and opportunity for breakthroughs in tumor immunotherapy.
